12 o'clock is pedal at the top of the circle. You'll probably have more luck fitting when the pedal is at the bottom of the circle (6 o'clock).
BTW, I don't think there's a right answer to your question, the angle of the thigh would depend upon your foot angle as well as the ratio of your calf-to-thigh-lengths.

Assuming you're legs are not abnormally short or long, I would use something around a 165 to 170mm crankset.
