Topically applied lipids / alternative to 'ORTHODOX' application?

  1. These two studies imply linoleic acid is an iron chelator / binder and
    effectively removes iron / hemosiderin from the skin .. ?

    This would mean topically applied lipids DO cross the skin .. ?

    Would anyone have any idea as to WHERE to find the AMOUNT of the lipids
    applied to the skin .. which .. would ENTER INTACT .. fully .. into the
    body .. ?

    If it has EVER been .. actually .. tested .. ?

    Linoleic acid (LA) is known to have a whitening effect on
    hyperpigmented skin, and is encapsulated in liposomes for topical
    application because of its low solubility in aqueous solution, although
    the effect of liposomalization of LA on the whitening activity has not
    been evaluated. In the present study, we evaluated the effect of
    liposomalization on the whitening activity of LA by using LA in
    ethanol, hydrogel containing LA, and hydrogel containing liposomal LA
    towards the UV-stimulated hyperpigmented dorsal skin of brownish guinea
    pigs. The whitening effect was far greater for hydrogel containing
    liposomal LA (0.1% w/w as a final concentration of LA) than for free LA
    in ethanol or hydrogel containing LA. Next, the whitening effect of LA
    was examined with UV-stimulated hyperpigmented human upper arm skin by
    using a hydrogel containing liposomal LA (0.1% LA) and non-liposomal LA
    (3.0, 10.0% LA). Liposomal LA (0.1%) showed a whitening effect
    comparable to 10.0% non-liposomal LA and was far more effective than
    3.0% non-liposomal LA. These results indicate that liposomal
    formulations are favorable for the transdermal application of LA.

    Experimental hemosiderosis: relationship between skin pigmentation and
    hemosiderin.

    Iron (totalling 7.5 mg per mouse in three doses) was injected into
    hairless mice to determine the relationship between skin pigmentation
    and hemosiderin deposition. The skin color reached its maximum 24 to 48
    hours after the last injection and then gradually faded over the
    subsequent 8 months. In the skin, hemosiderin granules were present
    extracellularly between collagen bundles as well as within dermal
    macrophages, Langerhans cells and indeterminate dendritic cells of the
    epidermis. A larger amount of iron was deposited in the facial than in
    the dorsal skin, resulting in darker pigmentation of the former. This
    study suggests that brownish discoloration of skin in hemochromatosis
    might be attributable in some degree to accumulation of hemosiderin and
    that pronounced hyperpigmentation of the face in hemochromatosis might
    be due to increased activation of melanocytes by a high content of
    hemosiderin.

  2. I have read that topically applied lipids can be used in patients on long
    term parenteral nutrition who cannot take intravenous lipids to prevent
    essential fatty acid deficiency. I do not know how much would cross the
    skin, but as lipids are obviously lipid soluble, some absorption would be
    expected.

  3. I previously said transdermal oils will penetrate the skin, but most
    references I have looked up on Pubmed suggest that the amount that reaches
    the body is limited, not enough to prevent essential fatty deficiency at
    least in patients on fat free parenteral nutrition

    Impact of topical oils on the skin barrier: possible implications for

    Topical therapy to enhance skin barrier function may be a simple,
    low-cost, effective strategy to improve outcome of preterm infants with
    a developmentally compromised epidermal barrier, as lipid constituents
    of topical products may act as a mechanical barrier and augment
    synthesis of barrier lipids. Natural oils are applied topically as part
    of a traditional oil massage to neonates in many developing countries.
    We sought to identify inexpensive, safe, vegetable oils available in
    developing countries that improved epidermal barrier function. The
    impact of oils on mouse epidermal barrier function (rate of
    transepidermal water loss over time following acute barrier disruption
    by tape-stripping) and ultrastructure was determined. A single
    application of sunflower seed oil significantly accelerated skin
    barrier recovery within 1 h; the effect was sustained 5 h after
    application. In contrast, the other vegetable oils tested (mustard,
    olive and soybean oils) all significantly delayed recovery of barrier
    function compared with control- or Aquaphor-treated skin. Twice-daily
    applications of mustard oil for 7 d resulted in sustained delay of
    barrier recovery. Moreover, adverse ultrastructural changes were seen
    under transmission electron microscopy in keratin intermediate
    filament, mitochondrial, nuclear, and nuclear envelope structure
    following a single application of mustard oil. Conclusion: Our data
    suggest that topical application of linoleate-enriched oil such as
    sunflower seed oil might enhance skin barrier function and improve
    outcome in neonates with compromised barrier function. Mustard oil,
    used routinely in newborn care throughout South Asia, has toxic effects
    on the epidermal barrier that warrant further investigation.

  7. They abstracts you cite only talk about fatty acids having effects upon the
    dermis. Whether the fat gets any farther than the dermis is not as well
    established. Certainly there seems to be some fatty acids making it into the
    central circulation, but most probably remains trapped in the dermis (which
    seems to be what you are looking for). Fats are mostly absorped and
    transported by the lymphatic system, and I do not think that there is much
    lymph drainage from the dermis.
