Wow you're brave. You'd better carry lots of water. Darwin in October is hot (mid 30s C) and humid, building up to the wet season. By the time you get 300km south to Katherine (and for most of the rest of the trip) it'll be dry and 35 - 40 deg C + daytime temperatures with large numbers of flies trying to land on your face, ears, legs, arms, in your mouth, nose.... The distances between towns/service stations as you get further south and into the Barkley Tablands (if you go via Queensland) can be 200km and more.
I've driven between Darwin and Sydney through outback QLD (the shortest route) many times. Not much traffic and huge distances, very isolated, hot, dry, dusty and mostly flat featureless grazing country. Some big trucks/road trains (ie truck with three trailers) and in Queensland the roads get rough and narrow. It's much more scenic to cross over to the coast of QLD via Townsville however thats about a thousand km extra distance and Sydney is still more than 2 thousand km south of there. Along the coast there will be much more traffic.
