I was thinking about creating an overlay of the route onto a map. Adding in succession layer per year which would then give an "average" depiction of the areas visited. From this it would be clear to see which areas haven't been visited. Also nice would be an option to select the years that you would be able to add (i.e. 1923 & 1955) for a comparison maybe.
Any interesing angles on this or techinical suggestions ?

No, but it sounds like a really lovely project. Read your Ed Tufte to warm up, and go to it!
I would suggest working with very thin lines for the route markings, at least compared to the typical tour maps you see right now.

This sounds like a very interesting project! Other people have had the same idea as yours in the past and even gone on to realize it, although not using modern GIS techniques. Paul Boury has written a 400 page book which gives a detailed analysis of the geography of the Tour de France, very interesting reading: http://www.editions-harmattan.fr/index.asp?navig=catalogue&obj=livre&no=10526
