Whenever I start to up the tempo a bit (while seated) the center of my right thigh feels as though it is clenching at the very top of the upstroke, just before applying downward pressure. This only happens on my right side. Any thoughts/similar experiences?

I occasionally experience some very minor strains in that area.
It's obviously not uncommon for a muscle strain to occur on only one leg.
It sounds to me like just that, some minor straining occurring. It makes sense, as the quads are at their most elongated state, and therefore the most stress, at the top of the pedal stroke because the knee is flexed quite a bit, and the quads are contracting.
It's possible your seat's a bit low, but if not, I've got some bad news for you: you're gunna have to do some STRETCHES😄
