Cool idea but have never seen it, but if you really want it try make it.
Idea # 1
Use new white handlebar tape. Get thin red fabric. With a stencil and a very sharp blade, cut circles smaller along the center of the tape. Then cut circle on the red fabric and use a fabric glue to glue the red fabric to the underneath of the white handle bar tape. Then wrap the tape on the bike. When I can't find stuff I want, I make it.
Idea # 2
You can try to use a red leather paint or stain on white handle bar tape and paint it on carefully using a stencil. I would make a indentation onto the leather to keep the red fluid from running all over the place. You might have to experiment with different stains or paints. Leather comes in tons of different colors and is usually colored on the leather.

You don't really see any considering all the climbers that normally wear the polka-dot jersey take of their handlebar tape.
If you want to dye/color the tape with polkadots.... I have considered using the hole reinforcements (like for notebooks paper) as templates for polkadots on projects. You could also punch holes in masking tape etc... then you could get unique and use other shapes by buying the appropriate punch at a craft store.
No polka dots, but Cinelli makes stripes and splashes. Maybe they will make polka dots if you suggest it to them. It's a good idea. I bet it would make hills much easier!
How about laying a piece of peg board that has the larger diameter holes over plain tape to use as a mask and just spray painting whatever color you like? You can even drill the holes out bigger if you like and they'll still be nice and evenly spaced. If you don't like the result hey, it's just handlebar tape, so you haven't lost much.
