A fastidiously cleaned and adjusted drivetrain helps for me 🙂 It's mainly psychological, but when everything meshes perfectly and the gears just click, things just seem to be easier.
I also got a significant improvement in comfort by putting on Vittoria Open Corsa CX tyres.. the ones I had before were nothing great (Vittoria Tecno Sport, and Conti Ultra 3000) but these are like magic. Longer rides feel like they're a couple dozen km shorter than what they used to be like.
They also feel as though they have less rolling resistance, but that might just be a placebo caused by feeling so much fresher on a ride.
They're not cheap in tyre terms, though you can get 'em for A$70 each (including an inner tube) from www.probikekit.com

I must agree with rek. The biggest effect (apart from set of lightweight wheels) I've noticed on how my bike rides and races is from tyre choice.
Currently I race and train on Michelin Pro Race, they just roll beautifully, and are extremely puncture resistant (bet you I'll puncture tomorrow now 🙂)
