if you pedal forwards, no.

Note that tandem stokers using conventional (non-tandem) cranks are
pedaling backwards, and so for them, yes.

Mostly. Thorn have an ingenious arrangement with Rohloff-equipped tandems
where the sync chain is on the right-hand side, using half of a
conventional double which also connects to the hub.

In theory now we have accepted enormous Q-factors there is no reason why
all tandems should have some funky quadrupeliser on the stoker cranks and
use this arrangement.

The thread actually moves in the crank when pedalling hard, so
threadlock will have no effect.

Oh, now that's not exactly true.

It will have the effect (as Werehatrack implied) of breaking your
teeth, your tools, your will, and/or your record of consecutive days
of not taking the Lord's name in vain in front of the neighbors' kids.

Yes, use grease. Unless, of course, you want to make it more
difficult removing the pedals. I use Loctite #242 (blue) in two
places - disk brake rotor bolts and the bolts that attach my cleats.
I see that Shimano brake calipers now come with some kind of white
threadlocker on the main bolt, so I guess maybe it's good to use it
there, too (but I never did before - and never had a problem). Any
other threads on a bike should be greased.
