This one did not have a dust cap and I unscrewed the screw as I said I flatten the tire and now when I try to pump it no air goes thru. I should be able to lock the pump in place then take both hands to pump the tire right? I tried the pump on my kids bike that have Schrader's and it works fine. The instruction state that it automatically adapts to Presta and Schrader.

OK, I wonder if you removed the Presta Valve from the valve stem, and not the lock nut at the end?

If the giant pump has two openings - one for schrader and one for presta - this is a common problem for this type of pump. Inside there is plastic floating ball that is press into one side of the outlet that automatically selects which opening air is to go out of. If you insert it on a presta valve, the ball is pressed against the schrader opening thereby closing that. The trouble is that the plastic ball sometimes get stuck into one side.

Try to shake out by knocking the air outlet on a hard surface before using. If that does not do it then you may have to open up air hose outlet.
