Our drug store which supply 90 days of medication has substituted
Glymepiride for Amaryl for my spouse to save on the cost of the
medication . Will there be any change in effectiveness of the generic
medicine as compared with brand drug ?

Possible, but not likely. I use generic when I can.
A generic drug has to have the same active ingredient(s)
as the name-brand drug, although the inert ingredients
may be somewhat different. There is usually some testing
to see that it works essentially the same, but nowhere
near the amount of testing of the name-brand drug.
