A tiny splinter of glass penetrated tire and tube last week. Fortunately I
knew enough to go looking for it before I fixed the flat. Just to be safe, I
made a boot from a dollar bill.
Now that the tire is back on and the new tube is inflated, I cannot find the
puncture in the tire.
So my question is, Should I replace this tire before my next big ride? Or is
a small puncture like this one functionally insignificant?

I think it's too small to worry about. There are almost certainly
other equally tiny and invisible cuts in the tire casing.

My rule of thumb, which has served me well for many years: Any tire
puncture damage that you can't find on casual inspection is not
important. Ride on.
