Can I run a 9 spd Shimano mtn cassette (something in the 34 tooth range) with a long cage rear derailleur even though I have Ultegra STI 10 spd shifters? I have a rather hilly century + ride coming up and want to save my knees some pain. I have a compact 50/34 and normally run a 12-27.

As much as I respect George's knowledge, I beg to differ on this one.
If its just for the one event, then its hardly worth swapping out to a triple if you have a MTB cassette/RD to spare.
10sp brifters will work with 9sp rears if you use the B Cable anchoring position:
